The metalloproteolytic activity of the anthrax lethal factor is substrate-inhibited.

چکیده
The anthrax lethal factor (LF) is a Zn2+ endopeptidase specific for mitogen-activated protein kinase kinases (MAPKKs), which are cleaved within their N termini. Here, the proteolytic activity of LF has been investigated using novel chromogenic MAPKK-derived peptide substrates, which allowed us to determine the kinetic parameters of the reaction. LF displayed maximal proteolytic activity at the pH and temperature values of the cell cytosol, which is its site of action. LF undergoes substrate inhibition, in keeping with the non-productive binding geometry of the MAPPK-2 N terminus to LF.

Potent inhibitors of anthrax lethal factor from green tea.
The anthrax lethal factor (LF) has a major role in the development of anthrax. LF is delivered by the protective antigen (PA) inside the cell, where it exerts its metalloprotease activity on the N-terminus of MAPK-kinases. PA+LF are cytotoxic to macrophages in culture and kill the Fischer 344 rat when injected intravenously. We describe here the properties of some polyphenols contained in green...
متن کاملThe cytotoxic activity of Bacillus anthracis lethal factor is inhibited by leukotriene A4 hydrolase and metallopeptidase inhibitors.
The lethal factor of Bacillus anthracis is central to the pathogenesis of anthrax. Its mechanism of action is still unknown. Recently, on the basis of sequence similarities, we suggested that lethal factor might act similarly to leukotriene A4 hydrolase (LTA4), a bifunctional enzyme also endowed with a metallopeptidase activity. Here we show that some inhibitors of the LTA4 hydrolase and metall...
متن کاملEvaluation the Efficacy of Anthrax Vaccine against Challenge with a Highly Virulent Strain of Bacillus anthracis Isolated from Soil in Sheep, Goats and Guinea Pigs in Iran
Protection of animals immunized against Bacillus anthracis is usually demonstrated by challenging with an appropriate dose of a strain of Bacillus anthracis that is lethal to unvaccinated animals inoculated at the same time. In this study the protective efficacy in anthrax vaccine (34F2 sterne strain spore) was evaluated in sheep, goats and guinea pigs challenged with subcutaneous inoculation...
متن کاملAsp 187 and Phe 190 residues in lethal factor are required for the expression of anthrax lethal toxin activity.
Anthrax toxin consists of three proteins, protective antigen, lethal factor, and edema factor. Protective antigen translocates lethal factor and edema factor to the cytosol of mammalian cells. The amino-termini of lethal factor and edema factor have several homologous stretches. These regions are presumably involved in binding to protective antigen. In the present study we have determined the r...
متن کاملConsequences and Utility of the Zinc-Dependent Metalloprotease Activity of Anthrax Lethal Toxin
Anthrax is caused by the gram-positive bacterium Bacillus anthracis. The pathogenesis of this disease is dependent on the presence of two binary toxins, edema toxin (EdTx) and lethal toxin (LeTx). LeTx, the major virulence factor contributing to anthrax, contains the effector moiety lethal factor (LF), a zinc-dependent metalloprotease specific for targeting mitogen-activated protein kinase kina...
